摘要
目的探讨口腔癌肿瘤细胞中c-erbB-2、bcl-2基因的表达水平及其在肿瘤形成、发展过程中的作用.方法采用荧光定量RT-PCR实验技术检测人舌鳞癌Tca8113细胞株中c-erbB-2、bcl-2基因mRNA表达水平.结果分光光度计检测纯度A260/A280均在1.8~2.0,通过基因表达扩增曲线图与看家基因GAPDH校正,得出目的基因的相对含量高.结论 bcl-2与c-erbB-2基因的表达水平在人舌鳞癌Tca8113细胞株中显著升高,可能在肿瘤形成、发展过程中起到一定的作用.
iv:Objective To investigate the expression of c-erbB-2,bcl-2 mRNA in human tongue carcinoma Tca8113 cell lines.To study mechanisms of tumor formation and progress. Methods To detect mRNA level of the c-erbB-2,bcl-2 genes in human tongue carcinoma Tca8113 cell lines with fluorescence RT-PCR.Results Detecting purity of c-erbB-2,bcl-2 mRNA with spectro- photometer A260/A280,it is between 1.8-2.0. We have obtained high relative content of purpose genes to rectify gene expression amplification diagram of curves with house-keeping gene GAPDH.Conclusion The level of c-erbB-2,bcl-2 genes is significant overexpression in human tongue carcinoma Tca8113 cell lines. c-erbB-2,bcl-2 genes have definite effect in the process of neoplasia and tumor development.
