摘要
目的:建立人血浆富马酸异丙吡仑检测的高效液相色谱方法。方法:血浆经高氯酸沉淀蛋白,以ZORBAX Eclipse XDB-C_(18)(4.6mm×150mm,5μm)为色谱柱;以乙腈-水-2%冰醋酸(20∶30∶50,V/V/V)为流动相,流速为0.9mL·min^(-1);检测波长为266nm。结果:富马酸异丙吡仑浓度在0.05-5.0mg·L^(-1)范围内线性关系良好(r=0.9999);最低检测限为0.05mg·L^(-1);高中低3个浓度的平均相对回收率和绝对回收率分别为(99.38±2.3)%和(85.58±3.8)%;日内、日间RSD分别为(3.6±2.3)%和(2.2±1.0)%。结论:方法简便、快速、准确可靠,适用于人血浆富马酸异丙吡仑浓度的测定及其药代动力学研究。
Objective;To develop a high performance liquid chromatography method for the determination of Isopropiram fumurate in human plasma. Methods: The plasma were precipitated by perchloric acid. The analytical column was packed with ZORBAX Eclipse XDB-C_(18)(4. 6 mm×150 mm,5 μm). The mobile phase consisted of acetonitrile-water-2% glacial acetic acid (20: 30: 50, V/V/V) with flow rate of 0. 9 mL·min^(-1). The UV detection wavelength was 266 nm. Results: Excellent line relationship was obtained in the range of 0. 05 mg·L^(-1) to 5. 0 mg·L^(-1) (r= 0. 9999), the determination limit of Isopropiram was 0. 05 mg L^(-1) and the average relative recoveries and absolute recoveries were (99. 38±2. 3) % and (85. 58±3. 8) % with intra-day RSD (3. 6±2. 3) % and inter-day RSD 2. 2±1. 0% respectively. Conclusion: The method is simple, rapid, accurate and can be used to determine the Isopropiram concentration in human plasma and for study of its pharmacokinetics.
