摘要
50年来抗生素在人类疾病治疗中发挥了重要作用 ,今后的几十年里它们也将是关键的治疗剂。尽管在过去的 2 0年中通过靶向筛选发现了一些微生物药物 ,但是这种筛选方法很难发现新类型药物。组合生物合成可以弥补这种不足 ,通过基因工程方法改造微生物基因和酶 ,产生新的抗生素 ,发现那些在自然界中不能发现的药物。
Antibiotics have been playing important roles in hu man therapeutics ov er the past fifty years. They are likely to remain crucial in the coming decades . Despite the fact that microbial metabolites are being found continually throug h targeted screening programs, fewer and fewer fundamentally new types of drugs have been found in this way during the past two decades. Combinatorial biosynthe sis seems to help change this situation through genetic engineering, the goal of which is to make new antibiotics by exploiting the microbial genes and enzymes that make these substances, thereby aiming to discover drugs that cannot be foun d in nature.
