摘要
随着高致病耐药菌的不断出现,临床上对新型抗生素的需要变得十分迫切,达托霉素的上市使得环脂肽类化合物成为抗生素研发的热点之一。本文综述了达托霉素的生物合成基因簇、生物合成途径以及基于组合生物合成原理的达托霉素结构改造工作,为进一步利用基因工程手段来合成新的达托霉素衍生物提供理论依据。同时也简单介绍了达托霉素的临床研究进展。
With the continual emergence of highly pathogenic drug-resistant bacteria,the clinical need for new antibiotics is becoming urgent.The marketing of daptomycin makes the cyclic lipopeptide compounds to be a hotspot for research and development of antibiotics.This review focuses on the progress of daptomycin gene cluster,biosynthesis pathway and structure modification of daptomycin by combinatorial biosynthesis.These would provide a theoretical basis for the biosynthesis of new daptomycin derivatives by genetic engineering.In addition,the clinical development of daptomycin is also presented briefly.
