摘要
目的探讨AB血清对人胚脑神经干细胞体外生长的作用,为神经细胞应用于临床,消除异蛋白污染提供依据。方法取妊娠12~16周合法人工流产胎儿大脑皮层,用H-DMEM培养液制备神经细胞悬液,分别接种于含有AB血清和马血清的H-DMEM培养液中。倒置显微镜下观察神经细胞的生长发育、增殖分化、细胞迁移、形态学变化等,免疫组织化学SP法鉴定神经细胞。结果含AB血清及马血清的神经细胞均能很好地生长,神经特异性烯醇化酶阳性出现时间无显著性差异(P>0.05)。结论AB血清可以用来培养神经细胞。使用AB血清培养神经细胞,具有消除异蛋白污染和动物血清带来的其它未知病毒污染的可能性,为今后神经细胞应用于临床提供依据。
Objective To explore the role of AB serum on the growth of human embryo cranial nerve stem cell in vitro, for applying nerve cell in clinic and precluding the contamination of alloabumin.Method Using the cerebral cortex of legally induced abortion fetus of 12-16weeks,nerve cell suspension in DMEM culture media was prepared and inoculated it into the DMEM culture media which contains AB serum and horse serum respectively.The growth and development, cell migration, morphology change and so on of the nerve cell under the invert microscope were observed, the nerve cells were characterizated by the method of immunohistochemistry streptavidin peroxidase(SP).ResultsThe nerve cell both in AB serum and horse serum could grow quite well and there were no significant differences between both the groups in the positive emergence time of specific enolase of the nerves (P>0.05).Conclusion Culturing nerve cell by AB serum could preclude the contamination of alloal-bumin,and eliminate the possibility of other unknown virus contamination with animal serum,and provide the basis of clinical applying in the future.[
