摘要
用根癌农杆菌感染芥菜型油菜子叶外植体,培养2~3d,然后转到含Km10~20mg/L的选择培养基上筛选转化体,在选择培养基上获得的Km抗性苗,进一步在含Km30~50mg/L的选择培养基上筛选3~4月,然后诱导生根,再生成完整植株.DNA分子杂交证明B.t.基因已整合到油菜基因组中,转化频率为1%~2%.
In this paper, we report the transformation of rape (Brassica Juncea).The cotyledons with the petioles cut from exenic seedings were infected with Agrobacterium tumefaciens strain AST 367 (pBS21 ), and cultured on MS selective medium containing 10 ̄20 mg/L kanamycin and 4. 5 mg/L 6-BA To select transformants. The transformed shoots were further subcultured on the selective medium containing 30 ̄50 mg/L kanamycin for 3 ̄4 months,and then rooted on MS medium containing 0. 5 mg/L IBA and 20 mg/L Km. The transformation frequency was about 1% ̄2%. Southern blot hybridization showed that some of the plants gave positive signal with insecticidal gene(B. t. gene)probe. The Plants were transplanted to soil in pots and developed to mature. seeds were obtained from self-pollinated transgenic plants.
