摘要
采用细胞生长曲线、细胞毒试验、克隆形成试验以及有丝分裂指数和多核率测定,观察了维胺酸(RⅡ)对人喉癌HEp-2细胞增殖的影响。结果表明:不同浓度RⅡ(2~6mg·L-1)接触细胞24h、48h和96h,HFp-2细胞增殖抑制效应与剂量呈正相关;与溶剂对照相比,当RⅡ浓度为2、4和6mg-L-1时,MTT法的细胞生长抑制率分别为10.83%、71.34%和82.17%(r=0.951,P<0.05),半数抑制浓度(IQ50)为3.01mg·L-1;克隆形成试验中RⅡ组细胞存活率分别为72.15%、49.32%和43.38%(P<0.01),呈现剂量相关性抑制(r=-0.967,P<0.05);RⅡ还抑制细胞有丝分裂指数和多核率(P<0.05或P<0.01),镜下可见癌细胞有形态学改变。表明RⅡ对人喉癌细胞的生长增殖具有抑制作用。
In our experiment,the effect of N-(4-carboxyphenyl)retinamde(RⅡ) on the proliferation of human laryngeal carcinoma cell line HEp-2 was observed by using the following methods:cell growth curves,cytotoxicity test,cell colony-forming test,mitotic index and multinuclear rate determination.The results showed that when the HEp-2 cells were treated with RⅡat the concentration of 2~6mg·L-1 for 24,48 of 96 hours,the inhibitory effect of R Ⅱwas in positive correlation with its concentration;when the concentrations of R Ⅱ were 2,4and 6 mg·L-1,the cell growth-inhibition rates induced by RⅡ in MTT assay,as compared with the solvent control,were 10.83%,71.34%and 82.17%,respectively(r=0.951,P<0.05).The half inhibition concentration(IC50) was 3.01 mg·L-1.In the cell coloy-forming test,the cell survival rates of various concentrations in the R Ⅱ group were 72.15%,49.32%and 43. 38%(P<0.01).The inhibition effect of R Ⅱ was in a dose-dependent manner(r=-0.967,P<0.05).R Ⅱ also depressed the mitotic index and multinuclear rate of the HEp-2cells(P<0.05)or P<0.01).Under microscopy,the morphologic changes induced by R Ⅱwere observed in HEp-2 cells.The results indicated that R Ⅱ possessed the inhibitory effects on the growth and proliferation of the human laryngeal carcinoma cell line HEp-2.
