摘要
研究了变性剂盐酸胍对几种蛋白质在高效疏水色谱中保留行为的影响。用液相色谱中溶质的计量置换保留模型和测流动相表面张力及蛋白质紫外吸收光谱方法研究的结果表明:在低浓度范围内,盐酸胍主要以疏水作用影响蛋白质的保留;而在高浓度区域内,盐酸胍的存在则显著影响蛋白质分子的构象,使其保留行为发生变化.
The retention behavior of five proteins in high performance hydrophobic interaction chromatography(HPHIC)under the mobile phase consisting of(NH_4)_2SO_4-KH_2PO_4 containing guanidine hydrochloride was investigated.By using the stoichiometric displacement model for retention(SDM-R)of solute in liquid chromatography,and the measuring surface tension of the mobile phase in the presence of GuHCl,also and UV spectra,it was found that in the range of low concentraction of GuHCl,the hydrophobic irnteraction is considered as the main contribution to the retention of the proteins,However,whem the concentration of GuHCl in the mobile phase is getting higher,the changes of the molecular conformation of proteins by means of GuHCl in the HPHIC become the most important factor to influecing the retention of proteins.
