抗阿霉素中国仓鼠卵巢上皮细胞的聚合微管蛋白和F-肌动蛋白的含量变化

摘要 运用显微光度术(MPVⅡ),研究抗阿霉素(ADR)的中国仓鼠卵巢上皮细胞(CHO)抗药株(RC1)的细胞骨架,同时测定单细胞的DNA和聚合微管蛋白相对含量、DNA和F-肌动蛋白相对含量,发现RCl聚合微管蛋白含量减低,F-肌动蛋白含量增加,并都与CHO有显著性差异。与CHO相比,RCl的F-肌动蛋白除含量增加外,排列较紊乱,并局部解聚,类似于转化细胞的形状。这表明,细胞骨架与多药抗药性有关,但是否直接或间接相关还有待于进一步研究。 On cytoskeleton in Adriamycin(ADR)-resistant Chinese ovary cell line(RCl)was researched by means of micro-fluorometry(MPVⅡ).The content of polymerized microtubulin and DNA were counted in single cell at the same time,and the content of F-actin and DNA were done by the same way.It is found that the total F-actin content of RCl displayed increasing expression in comparision with that of CHO,while the total polymerized microtubulin decreased in RCl.F-actin,in addition to increase in the content in comparision with CHO,was in disorder,and dissocated partily like transformed cells.This shows that cytoskeleton is correlated with multidrug resistance though it is unknown whether they are related with each other directly or indiretly.
