摘要
用日本血吸虫成虫特异性抗原(sj-SAAg)和虫卵特异性抗原(sj-SEAg)免疫家兔制备免疫血清,继用SephadexG200柱层析法提取家兔免疫血清中抗sj-SAAg和抗sj-SEAg的两种特异性多克隆抗体。将人“O”型血红细胞分别与上述两种抗体结合制备红细胞—抗体结合物(RBC-Abs),并研究了该结合物的应用方法。初步研究结果表明,实验所用载体以混合纤维素膜最佳,检测31例急性血吸虫病人阳性率为96.8%,20例正常人假阳性率为5%。
In the present studies firstly, the specific antigens in adults (Sj-SAAg) and eggs (Sj-SEAg) were used to immunize rabbits for preparing immuno-sera, then, the polyclonal antibodies against Sj-SAAg and Sj-SEAg were respectively isolated from the immuno-sera by Sephadex G200 Chromatgraphy, finally, a conjugates of RBC-Abs were prepared by the combination of human type "O'' erythrocytes with the antibodies against Sj-SAAg or Sj-SEAg and the method using the conjugates as marker was studied. The preliminary study shows that mixed cellulose ester is best carrier for this detecting method. 31 acute schistosomiasis patients and 20 normal controls diagnosed by this method, positive rates are 96.8% and 5%, respectively.
