胃粘膜疾病组织芯片制作及13种细胞周期调控因子的检测 被引量:1

摘要 目的:探讨制作胃粘膜疾病组织芯片的方法学及组织芯片技术的优缺点。方法:采取免疫组织化学技术检测胃粘膜疾病组织芯片中13种细胞周期调控因子(Ubipuitim、P16、P21、P27、P53、P57、chclin A、chclin B、chclinD1、cyclin E、CDK2、CDK4、CDK6)表达情况。结果:肠型胃癌中,Ubipuitin的表达高于慢性浅表性胃炎和肠化生,P16、F27的表达低于慢性浅表性胃炎和肠化生,P21、P57的表达低于慢性浅表性胃炎,P53的表达高于慢性浅表性胃炎、肠化生和异型增生,cyclin A、cyclin B、cyclinD1、cyclin E、CDK2、CDK4、CDK6的表达高于慢性浅表性胃炎和肠化生。结论:采用病变组织芯片方法检测可在最短的时间内获取病变组织13种细胞周期调控因子的表达情况,且节约研究经费、降低劳动量,具有重要的科研和应用价值。 Objective:To explore the methodology of histological chip making in gastric mucosa diseases and determine the advantage and disadvantage of the technique. Method: The expression of 13 cell cycle modulators in the chip were delected by . immunohistochemical technique in patients with gastric mucosa diseases. Results: In intestinal gastric carcinoma, expression of U-bipuitin was higher than that in chronic superficial gastritis and intestinal metaplasia, Expression of F16 and P17 was lower than chronic superficial gastritis and metaplasia,Expression of P21 and P57 lower than chronic superficial gastritis;Expression of P53 higher than chronic superficial gastritis, metaplasia and heterotypical hyperplasia; Expression of cyclin A, cyclin B, cyclin D1 , cyclin E, CDK2 and CDK6 was higher than chronic superficial gastritis and metaplasia. Conclusion: The expression of 13 cell cycle modulators can be detected by histological chip technique in short time. It is cost efficient and has important value for scientific research.
