摘要
目的 :建立高效液相色谱 -荧光检测法测定染色虾米中的碱性玫瑰精含量的检测方法。方法 :样品经均质 ,无水乙醇 -氨水 -水 (7+ 2 + 1)溶液浸提 ,SPE -HLB柱固相萃取 ,用LunaODSC18分离柱 ,甲醇 + 0 4g/L十六烷基溴化铵的水溶液 (70 + 3 0 )为流动相 ,荧光检测波长 :Ex为 5 5 4nm ,Em为 5 70nm。结果 :玫瑰精线性范围为 0 0 5~10 0 0 μg/mL,r2 =0 999,平均回收率为 85 92 % ,RSD =4 63 % (n =6) ,最低检出限为 2 μg/kg。结论 :该方法灵敏度高 ,重现性和选择性好 ,易于操作 ,是检测染色虾米中碱性玫瑰精含量之有效定量方法。
Objective:To establish a HPLC method to measure Rhodamine B content in driedshrimp Methods:The sample was processed by homogenized,ethanol-ammonia-water(70+20+10)solution extraction,the extraction was cleaned-up and concentrated by SPE on a HLB column,the chromatography was performed by reversed-phase HPLC on a C 18 column with fluorescence detection at Ex: 554 nm ,Em: 570 nm ,the mobile phase was methanol- 20 mmol/L ammonium acetate(60:40).Results:The linear range was 0 05~ 10 00 μg/ml , r 2=0 999,the average rate of recovery was 85 92%,RSD=4 63%( n =6),the detection limit was 2 μg/kg Conclusion:The method was efficient in detecting Rhodamine B with high sensitivity,good reproducibility and selectivity,simple operation [
