摘要
目的 研究乳腺癌组织和细胞株中黑色素瘤抗原基因 (Mage)的表达情况 ,为乳腺癌的免疫治疗提供依据。方法 采用RT PCR法检测 3 3例乳腺癌组织和 4株常用乳腺癌细胞株MCF 7、Sk Br 3、MDA MB 43 5s和TM 40D中Mage基因的表达情况。结果 3 3例乳腺癌组织中 9例 (2 7.3 % )至少表达 1种Mage A基因 ,4例 (12 .1% )Mage A 1阳性 ,7例 (2 1.2 % )Mage A 3阳性 ,细胞株MCF 7和Sk Br 3中Mage A1和Mage A 3呈阳性表达 ,MDA MB 43 5s和TM 40D中Mage A 3呈阳性表达。 结论 Mage基因在乳腺癌中表达率较高 ,Mage蛋白有望成为乳腺癌免疫治疗的靶抗原。
Objective To study the expression of Mage-A1、A3 in breast cancer tissue and four breast cancer cell lines and to explore the feasibility of immunotherapy with Mage gene encoding proteins in breast cancer.Methods The expression of Mage-A1、A3 in breast cancer tissue and four breast cancer cell lines,including MCF-7、Sk-Br-3、MDA-MB-435s and TM40D,was detected by reverse transcription polymerase chain reaction(RT-PCR).Results The positive expression rate of Mage-A in breast cancer tissue was 27.3% with 4(12.1%) cases of Mage-A1(+) and 7(21.2%) cases of Mage-A3(+).Both Mage-A1 and Mage-A3 were positive in breast cancer cell line MCF-7 and Sk-Br-3.MDA-MB-435s and TM40D expressed Mage-A3.Conclusion Mage genes has a high expression rate in breast cancer.Gene encoding proteins may be used as Mage-peptide-based active immunotherapy.
