摘要
目的 了解舟山眼镜蛇毒细胞毒素 - (CTX- )的抗肿瘤作用。 方法 应用磺胺邻二甲氧嘧啶(SRB)法观察不同浓度 CTX- 对人肝癌 Bel74 0 4细胞和人早幼粒白血病 HL - 6 0细胞的生长抑制作用。观察CTX- 0 .4 ,0 .2 ,0 .1mg/ kg腹腔注射对 U 14荷瘤小鼠瘤块质量的影响并计算抑瘤率 ;CTX- 0 .4和 0 .2 m g/kg静脉注射对艾氏腹水癌 (EAC)小鼠腹水形成的影响并计算腹水减少率。 结果 CTX- 可明显抑制人肝癌Bel74 0 4细胞和人早幼粒白血病 HL - 6 0细胞的生长 ,其 IC50 分别为 (1.12± 0 .13)和 (1.6 1± 0 .18)μg/ m L。 CTX- 0 .4 ,0 .2和 0 .1m g/ kg腹腔注射× 10 d,对宫颈癌 U14抑瘤率分别为 30 .5 0 % (P<0 .0 1) ,19.0 2 % (P<0 .0 5 )和12 .0 8% (P>0 .0 5 ) ;CTX- 0 .4和 0 .2 mg/ kg静脉注射× 3d对 EAC小鼠腹水形成减少率分别为 2 5 .5 % (P<0 .0 5 )和 14 .5 % (P>0 .0 5 )。 结论 舟山眼镜蛇毒细胞毒素 - 具有明显的抗肿瘤作用。
Objective To investigate the antitumor effects of cytotoxin-ⅩⅡ(CTX-ⅩⅡ) isolated from the venom of Naja atra. Methods The inhibition effects of cytotoxin-ⅩⅡ on the cultured Bel7404 cells and HL-60 human leukemia cells were observed by sulforhodamine B(SRB) method. Uterus cervix cancer U14 bearing mice was treated with CTX-ⅩⅡ(0.4, 0.2 and 0.1 mg/kg, ip). Ehrilich ascites carcinoma cells(EAC) bearing mice was treated with CTX-ⅩⅡ(0.4 and 0.2 mg/kg, iv). Results The cytotoxin-ⅩⅡ inhibited the cultured Bel7404 cells and HL-60 human leukemia cells, IC 50 of CTX-ⅩⅡ on Bel7404 cells and HL-60 human leukemia cells was (1.12±0.13) and (1.61±0.18) μg/mL, respectively. CTX-ⅩⅡ 0.4, 0.2 and 0.1 mg/kg, ip×10 d inhibited the growth of U14 in mice were 30.50%(P<0.01), 19.02%(P<0.05), 12.08%(P>0.05); CTX-ⅩⅡ 0.4, 0.2 mg/kg, iv×3 d also showed regression of ascetic tumors(EAC) in mice 25.5%(P<0.05) and 14.5% respectively(P>0.05). Conclusion cytotoxin-ⅩⅡ exhibited significant antitumor effect in cultured tumor cell or in tumor-bearing mice.
