缺氧诱导成骨细胞VEGF的表达及其对成骨细胞分化的影响 被引量:1

摘要 目的:研究在缺氧条件下体外培养成骨细胞诱导血管内皮生长因子(VEGF)的表达,及其对成骨细胞分化 的影响。方法:酶消化法传代培养乳SD大鼠颅盖骨细胞并建立缺氧模型,采用激光共聚焦显微镜检测VEGF的表 达;骨钙素放射免疫法检测骨钙素含量。结果:在培养的前3d,缺氧组细胞VEGF的光密度值及骨钙素含量均高于 对照组。结论:缺氧(1~3d)诱导成骨细胞VEGF的高表达并促进成骨细胞的分化。 Objective: This study is to investigate the expression of VEGF as well as differentiation after exposure of osteoblasts to hypoxic stimulus.Methods: The osteoblasts obtained from the newly born calvaria of SD rats were cultured by the method of enzyme digestion in vitro.After establishing the hypoxic model, we detected the expression of VEGF by laser confocal microscope and explored the OCN content by radioimmunoassy. Results:When the cells were cultured for the first three days , the O.D. value of VEGF and the OCN content of hypoxic group were increased comparing with that of normal group. Conclusions: Hypoxia(1~3 days)can increase the high expression of VEGF and improve the differentiation of osteoblasts.
