摘要
目的:鉴于脏器功能障碍直接影响疾病的预后的生活质量,其损伤后的再修复尤为重要。因此,探讨β-七叶皂甙钠(sodiumaescinate,SA)对胰腺缺血再灌注损伤(ischemia-reperfusion,IR)的保护作用及可能机制。方法:实验于2002-12/2004-01在徐州医学院神经生物学研究中心实验室和解放军第九十七医院实验科完成。40只大鼠随机分为5组:假手术组,IR1h组,IR6h组,SA+IR1h组,SA+IR6h组。测定血淀粉酶,脂肪酶含量;光镜观察胰腺组织结构改变;检测胰腺组织丙二醛含量和超氧化物歧化酶(superoxidedismutase,SOD)活性;测定胰腺组织干湿比。结果:①SA+IR1h组淀粉酶、脂肪酶含量较IR1h组显著降低(P<0.05),SA+IR6h组较IR6h组降低更明显(P<0.01)。②与IR1h组、IR6h组相比,同期SA+IR1h组、SA+IR6h组组织病理学评分降低。③IR组与同期SA+IR组相比,SOD活性差异无显著性意义(P>0.05)。SA+IR1h组较IR1h组丙二醛含量显著降低(P<0.05),SA+IR6h组较IR6h组丙二醛含量降低更明显(P<0.01)。④SA+IR1h组与IR1h组间胰腺干湿比减少差异无显著性意义。SA+IR6h组较IR6h组胰腺干湿比减少显著改善(P<0.05)。结论:SA对胰腺IR损伤有保护作用,其保护作用可能与其抑制IR后胰腺氧自由基的形成和减轻组织水肿有关。
AIM:To investigate the protective effects of sodium aescinate(SA) on the rat pancreatic ischemia reperfusion(IR) injury and its possible mechanism,based on that the visceral dysfunction can affect the prognostic quality of life directly and the repair is very important after injury. METHODS:The experiment was conducted in the Laboratory of Research Center for Neurobiology,Xuzhou Medical College and Department of Experimental Medicine,the 97 Hospital of Chinese PLA from December 2002 to January 2004.Forty male Spraque Dawely(SD) rats were randomly divided into 5 groups:sham group,1 hour IR group,6 hour IR group,SA+ 1 hour IR group and SA+ 6 hour IR group.The plasma concentrations of amylase and lipase were measured,and the pathological and ultrastructural changes of pancreatic tissue were observed under light microscope.The level of malondialdehyde(MDA) and activity of superoxide dismutase(SOD) in pancreatic tissue were determined.Pancreatic dry weight(DW) to wet weight(WW) were examined. RESULTS:① The plasma concentrations of amylase and lipase were significantly lower in SA+ 1 hour IR group than in 1 hour IR group(P< 0.05),and much significantly in lower SA+ 6 hour IR group than in 6 hour IR group(P< 0.01).② The histological scores in SA+ 1 hour IR group and SA+ 6 hour IR group were significantly lower compared with those in 1 hour IR group and 6 hour IR group respectively.③ The activity of SOD in IR group and SA+ IR group were not significantly different(P >0.05).The MDA level of SA+ 1 hour IR group and SA+ 6 hour IR group were significantly lower than those of 1 hour IR group and 6 hour IR group respectively(1 hour IR,P< 0.05;6 hour IR,P< 0.01).④ The ratio decrease of DW to WW of pancreatic tissue in 1 hour IR group were insignificantly different from SA+ 1 hour IR group.Compared with 6 hour IR group,the ratio decrease was significant in SA+ 6 hour IR group(P< 0.05).
