摘要 目的 评价自体外周血造血干细胞(APBSC)的动员、采集及冻存方法。方法 18例病例中,恶性淋巴瘤 16例(NHL14例,HD2例),睾丸肿瘤 2例。病人行诱导化疗完全缓解后,采用大剂量联合化疗加小剂量粒细胞集落刺激因子(G-CSF)进行外周造血干细胞动员。用CS-3000,Plus血细胞分离机采集外周血造血干细胞,将分离的单个核细胞 (MNC)经程序降温仪降至 -80℃后,冻存 -196℃液氮。冷冻前及解冻后分别行MNC计数和粒单系祖细胞集落(CFU-GM)培养。结果 动员后外周血WBC及MNC总数明显增加,与动员前比较,差异有显著性。冷冻前后,MNC计数,CFU-GM集落数无明显差异。结论 大剂量联合化疗加小剂量G-CSF动员方案是安全有效的。冷冻保存APBSC及复苏过程对细胞损伤较小,值得推广使用。 Objective To Evaluate the methods of mobilization, collection and freezen store of autologous peripheral blood stem. Methods There were 16 cases malignant lymphoma(14 cases NHL; 2 cases HD) and 2 cases neoplasm. When 18 patients received mitigation after neoadjuvant chemotherapy, we used huge dose combined chemotherapy plus lower dose G-CSF to mobile peripheral blood. CS-3000 and PLUS blood cell separate machine were used to collect peripheral blood stem cell. Procedure Lower Temperature instrument was used to lower MNC to subzero 80 celsius degree and stored in subzero 196 celsius degree liquid nitrogen. Before freezen store and after unfreezen store, we counted MNC and cultured CFU-GM. Results The WBC and MNC counts of peripheral blood were significantly increased, compared with the counts of WBC and MNC before mobilization. The difference was significant. There was no difference in the counts of MNC and CFU-GM group before and after freezen store. Conclusion Huge dose combined chemotherapy plus lower dose G-CSF mobilization regimen are safe and effective. With freezen store of APBSC and recovery, cell damage are small.This method should be worthy of being promoted.
