摘要
目的 :检测低温缺氧 /复温供氧新生鼠出血肺组织中缺氧诱导因子 1α(HIF 1α)的表达。方法 :80只Wistar新生大鼠随机分为对照组 (C组 )和三个实验组 (H2 、H4、H4R2 组 ) ,H2 和H4组分别予以单纯低温 (10℃ )缺氧 (5 %O2 ) 2、4h ,H4R2 组低温缺氧 4h后 ,再暴露于常温 (37℃ )高氧 (95 %O2 )环境 2h。采用免疫组化 (SP法 )检测肺组织HIF 1α蛋白表达水平。结果 :H2 、H4组HIF 1α表达水平显著升高 (与C组比较 ,P <0 .0 5 ) ,H4组则有显著下降 (与H2 、H4组比较 ,P <0 .0 5 ) ,但仍高于C组。低温缺氧阶段其表达水平与低温缺氧时间、肺损伤程度呈正相关 (r=0 .72 9、0 .76 ,P <0 .0 1)。结论 :HIF 1α参与了低温缺氧 /复温供氧诱导的新生大鼠肺出血病变过程。
Objective:To explore the expression of Hypoxic inducible factor-1α (HIF-1α) protein in pulmonary tissue of newborn rats with pulmonary hemorrhage (PH) induced by hypothermia-hypoxia/rewarming-reoxygenating. Methods Eighty newborn rats were randomly divided into control group (group C) and three experiment groups(group H 2, H 4, H 4R 2). Group H 2 and H 4 were exposed to hypothermia(10 ℃)-hypoxia(5% O 2) for 2 or 4 hours respectively; Group H 4R 2 was in hypothermia-hypoxia for 4 hours and then placed into hyperthermia(37 ℃)-hyperxia (95% O 2) for 2 hours. The HIF-1α protein were detected in pulmonary tissue by immunohistochemical method. Results: The levels of HIF-1α expression increased significantly in groups H 2 and H 4 (compared with group C, P<0.05), declined dramatically in group H 4R 2(compared with group H 2, H 4).The HIF-1α expression were positively correlated with the time of hypothermia-hypoxia, and with PH degree in hypothermia-hypoxia phase (r=0 729,0.76, P<0.01). Conclusion HIF-1α may play a role in pathologic process of PH induced by hypothermia-hypoxia/rewarming-reoxygenating.
