摘要
为探讨c-erbB-2基因在乳腺癌发生早期的作用,用免疫组化方法检测36例乳腺单纯性增生、31例不典型增生、30例乳腺癌中p185蛋白的表达,用狭缝印迹法检测上述组织中c-erbB-2基因的扩增。结果显示p185 蛋白在单纯性增生、不典型增生,乳腺癌中的表达率分别为0%,25.8%(8/31),56.7%(17/30);c-erbB-2基因的扩增率分别为0%,19.3%(6/31),36.7%(11/30)。表明c-erbB-2基因扩增发生于不典型增生阶段,在增生病向乳腺癌进展中起重要作用,可作为预测乳腺癌的辅助分子标记物。
In order to investigate the role of c-erbB-2 gene in the early stage of human breast carcinogenesis, p185 protein expression was detected by using S-P immunohistochemical technique in 36 cases of simple hyperplasia of mammary glands, 31 cases of atypical hyperplasia of mammary glands, 30 cases of breast carcinomas, and c-erbB-2 amplification was determined by DNA slot blot hybridization in their tissues. Expression of p185 protein was found in 25.8%(8/31) of breast atypical hyperplasia and 56.7%(17/30) of breast carcinomas, and c-erbB-2 amplification was observed in 19.3%(6/31) of breast atypical hyperplasia and 36.7%(11/30) of breast carcinomas. No expression and amplification of c-erbB-2 were found in simple hyperplasia. It indicated that amplification was involved in breast carcinogenesis, which may play an important biological role in the development of breast cancer from atypical hyperplasia, and could be a potential molecular marker for the prediction of breast carcinomas.
