摘要
目的: 研究人脐静脉内皮细胞 (HUVEC)感染汉坦病毒(Hantavirus, HTV)后应激反应的规律及意义。方法: 采用免疫细胞化学染色法和核酸分子原位杂交, 检测热休克蛋白 70(HSP70)的表达; 用RT --PCR观察HSP70mRNA水平变化的规律。结果: HTV感染HUVEC后, 免疫细胞化学染色法可检出HSP70呈高表达, 原位杂交发现细胞质内有HSP7mRNA的阳性信号。感染后不同时间点RT- PCR的结果表明与对照组相比较, HSP70mRNA的水平在感染后迅速升高并持续高表达 (P<0. 05 )。结论: HTV可诱导HUVEC高表达HSP70。HSP70可能具有抑制病毒复制和保护内皮细胞的作用。
AIM: To study stress response in human umbilical vein endothelial cells(HUVEC) infected with Hantavirus (HTV). METHODS: The expression of HSP70 was detected by immunocytochemical staining, in situ nucleic acid hybridization and RT-PCR. RESULTS: After being infected with HTV, the expression of HSP70 in HUVEC increased. RT-PCR showed that the level of HSP70 mRNA increased rapidly and kept at a high level in comparison with the normal control(P<0.05). CONCLUSION: The over-expression of HSP70 in HUVECs can be induced by Hantavirus, which may play a role in the inhibition of virus reproduction and the protection of cells from virus infection.
