摘要
【目的】探讨海洛因成瘾大鼠心肌超微结构及心肌凋亡的改变。【方法】建立大鼠海洛因成瘾模型,采用透射电镜及脱氧核苷酸末端转移酶介导的缺口末端原位探针标记(TUNEL)观察心肌超微结构改变及心肌凋亡的情况。【结果】大鼠海洛因成瘾组电镜改变主要表现在核浓缩,核变小,核膜皱缩,染色质凝集成块,线粒体嵴排列紊乱、消失及空泡变等。TUNNEL检测结果显示海洛因成瘾大鼠心肌出现凋亡阳性细胞。【结论】上述改变说明海洛因对心肌可造成损害。
To explore the changes of myocardial ultrastructure and to detect the apoptosis cells in heroin dependence in rats. Heroin addict model were built in six Sprague-Dawley(SD) rats, Electronic microscope and terminal deoxynucleotidyl transferase-mediated biotin-dUTP nick-end labeling(TUNEL) were used for investigate the changes of myocardial ultrastructure and apoptosis. Electronic microscope shows that structural of many nuclear abnormalities such as nuclear concentrate or reduce, nuclear membrane shrink, chromatin agglutinate. Many mitochondria abnormalities such as cristal disorder, disappear or hollow, et al. Positive staining of myocardial apoptosis cells had been detected by the way of TUNEL. [Conclusion] Above-mentioned changes indicated that heroin could injure myocardium.
