摘要
血管生成是组织再生的必要条件,促进血管生成技术目前已经成功地开始应用于缺血性疾病的治疗,而且正在成为组织工程研究的前沿。治疗性血管生成要求血管生成因子能够在缺血区域中以持续、稳定、局限的低剂量释放,而生物材料作为因子的缓释载体将有效的解决此难题。这些生物材料对调节因子的稳定、活性、释放、分布等有着重要作用。来源于天然材料的包括:胶原与明胶、纤维蛋白、藻酸盐、壳聚糖、琼脂糖等;而来源于人工合成的材料主要为:聚乳酸与聚羟基乙酸的共聚物、聚乙二醇水凝胶以及其它可吸收材料。新的改良材料不仅可以维持因子活性,而且其有效释放将与局部组织愈合再生过程相协调。
Angiogenesis is essential to tissue regeneration.Technology to promote angiogenesis has now begun to be successfully used in the treatm ent of ischemia diseases and has beco me one of the most active areas of tiss ue engineering.Therapeutic angiogen esis indicates a need for continuous local delivery of low doses of angiog enic activ-ities at the target site.A variety of natural or synthetic polymers have b een employed as carrier devices for a ngiogenic growth factors and believed to solve the problem efficiently.They play a highly functional role in modulatin g the sta-bility,activity,release and spatial localization of these factors.Na tural matrices include alginate,agarose,chitosan,fibrin,collagen and gelatin.Synth etic materials include poly(lact ide-co-glycolide),poly(ethylene glycol)hydrogel matrices and some other absorbable o nes.With development of the technology,the newly improved materials not only protect growth factor activities,b ut also achieve optimal delivery with coalescence and regeneration of lo cal tissue.[
