富硒蚕蛹氨基酸诱导人肝癌细胞SMMC-7721凋亡的研究 被引量:12

摘要 目的 研究富硒蚕蛹氨基酸对人肝癌细胞SMMC 772 1的作用。方法 以人肝癌细胞SMMC 772 1为模型 ,采用原子荧光分光光度计测定蚕蛹硒含量 ,通过噻唑蓝(MTT)比色法检测细胞生长及活力 ,用倒置荧光显微镜观察细胞形态学变化 ,流式细胞仪研究细胞的凋亡及细胞周期变化。结果 ①陕西紫阳县蚕蛹中硒的含量为普通蚕蛹硒含量的 2 15倍 ,②富硒蚕蛹氨基酸在 0 5、1 5和 2 5 μmol·L-1硒浓度下能显著性抑制肝癌细胞生长 ,并具有浓度和时间依赖性。亚硒酸钠对细胞生长的抑制作用不如富硒蚕蛹氨基酸。相反 ,普通蚕蛹氨基酸促进细胞的生长。③富硒蚕蛹氨基酸作用于细胞后 ,倒置荧光显微镜观察到细胞形态学变化及细胞存活率降低。④流式细胞仪检测到 0 5、1 5和2 5 μmol·L-1富硒蚕蛹氨基酸所致细胞凋亡及细胞周期的变化 ,该氨基酸能阻滞SMMC 772 1的细胞周期于G2 /M期 ,使G2 /M期细胞比例增加 ,而G0 /G1期细胞比例下降。结论 富硒蚕蛹氨基酸能诱导SMMC 772 1细胞的凋亡 ,其抗癌机制可能与其改变细胞周期进而诱导细胞凋亡相关联。 Aim To study the effect of selenium(Se)-abundant a mi no acids from Ziyang silkworm pupa on human hepatoma cells. Methods H uman hepatoma cell line SMMC-7721 was selected as the model for study. Se-abun dant amino acids were prepared from Se-abundant silkworm pupas. Se contents wer e detected by atomic fluorescence spectrometer. Cell growth and viability were m easured by MTT method. Cellular morphology was observed by inversed fluor escence microscope. Apoptosis and cell cycle changes were determined by flow cytometry. Results ①The selenium content in Ziyang silkworm pupa was 215 times of that in normal silkworm pupa;②Se-abundant amino acids at final Se concentr ations of 0.5,1.5 and 2.5 μmol·L -1 significantly inhibited hepatoma cell growth and proliferation. The inhibitory effect was dose and time dependen t. Normal amino acids from normal silkworm pupa prompted cell gro wth. Sodium selenite at thesame concentrations only slightly inhibited cell growth;③ Se-abundant amino acids significantly and dose-dependently induced cell morphological changes and decreased survival cell rates;④Flow cytometry analysis confirmed the fact that Se-abundant amino acids at 0.5,1.5 and 2.5 μmol·L -1Se significantly and dose-dependently induced hepatoma cell apoptosis, increased G 2/M fraction and decreased G 0/G 1 fraction in the cell cycle. Conclusions Se-abundant amino acids could inhibit cell growth and induce apoptosis of human hepatoma cell SMMC-7721. The anti-hepatoma mechanism of Se-abundant amino acids is related to the changes of cell cycle.
