摘要
目的 研究诱导热休克蛋白 (HSP 70 )高表达对射线引起的前体血细胞凋亡的保护作用。方法 前体血细胞分别在41℃温浴 2h和加入砷酸钠诱导HSP 70 ,然后射线辐照。对照前体血细胞则只是射线辐照而不预热。并用HSP 70反义寡核苷酸链抑制HSP 70合成 ,进一步观察HSP 70与细胞凋亡的关系。利用DNA碎片检测法检测细胞凋亡 ,HSP 70的量利用WesternBlotting检测。结果 预热和砷酸钠都可以诱导前体血细胞HSP 70高表达 ;预热同时可以增加细胞凋亡的基础量 ,HSP 70高表达可以明显减少辐射诱导的细胞凋亡。在预热前导入HSP 70反义核酸链到前体血细胞 ,就可以减少HSP 70的抑制细胞凋亡的作用。结论 不论何种因素诱导的HSP 70高表达都可以保护前体血细胞因为辐射引起的细胞凋亡。
[Objective] To study the protective function of HSP 70 over expression for the precursor blood cells against radiation-induced apoptosis. [Methods] The precursor blood cells were either preheated in water of 41℃ for 2 hours or affected with sodium arsenate so as to induce HSP 70, then the cells were irradiated. The control cells were only irradiated but not preheated. Then HSP 70 antisense oligonucleotide was used to restrain the composition of HSP 70, then we proceeded to examine the relationship between HSP 70 and cell apoptosis. DNA fragmentation method was used to test the apoptosis of cells; Western Blotting was used to test the amount of HSP 70. [Results] Both preheating and sodium arsenate can induce the HSP 70 over expression, which can significantly reduce radiation-induced apoptosis. Introduction of HSP 70 antisense oligonucleotide prior to preheating can reduce the protective function of HSP 70. [Conclusion] HSP 70 over expression induced by whatever factors can protect the precursor blood cells against radiation-induced apoptosis
