摘要
目的探讨脑缺血-再灌注损伤中肿瘤坏死因子α(TNF-α)、白细胞介素-1β(IL-1β),白细胞介素-6(IL-6)水平的变化及丙泊酚对它们的影响。方法72只成年Wistar大鼠随机分为三组:A组为假手术组(n=8);B组为对照组(n=32),于缺血前10min腹腔注射生理盐水10 ml/kg,据实验要求再均分为4个亚组,均缺血2 h,再分别灌注1、6、12、24 h;C组为丙泊酚组,于缺血前10 min腹腔注射丙泊酚100 mg/kg,亚组分组同B组。观察血中TNF-α、IL-1β、IL-6的浓度变化和血中丙二醛(MDA)、超氧化物歧化酶(SOD)的变化以及光镜下缺血大脑组织病理改变。结果 丙泊酚组的TNF-α、IL-1β、IL-6、MDA含量明显低于对照组(P<0.05),而SOD活性高于对照组。光镜结果提示,丙泊酚组局灶性脑缺血-再灌注损伤轻于对照组。结论丙泊酚抑制脑缺血-再灌注时炎性细胞因子的表达,从而减轻该类细胞因子介导的炎性损伤,具有一定的脑保护作用。
Objective To explore the effect of propofol on plasma tumor necrosis factor alpha (TNF-α) ,interleukin-1β (IL-1β) and interleukin-6 (IL-6) in rats following cerebral ischemia-reperfu-sion injury. Methods Focal cerebral ischemia modle was induced with occluding the middle cerebral artery (MCA) . Seventy two adult Wistar rats were divided randomly into three groups. Sham operation was done in group A. The control group (group B) received brain ischemia for two hours,and was subdivided into four groups according to the reperfusion time of 1,6,12,or 24 h respectively. Normal saline, 10 ml/kg, was injected intraperitoneally before ischemia. Propofol group (group C) was given propofol, 100 mg/kg instead of normal saline as in group B. The serum concertrations of TNF-α,IL-1β,IL-6 and malondialdehyde(MDA) ,and activities of superoxide dismutase (SOD) were measured. Pathological examination of the cerebral tissue was carried out. Results Blood levels of TNF-α,IL-1β, IL-6 and MDA of group C were significantly lower than those of group B(P<0. 05), but SOD was markedly higher in group C than that in group B. Under microscope, the cell structure was still perfect in group C, whereas that in group B was swelling, karyorrhexis, karyolysis. Conclusion The expression of the inflammatory factors is suppressed by propofol with less cerebral ischemia-reperfusion injury.
