摘要
目的 :研究白介素 -1β(IL -1β)对膀胱癌细胞基质金属蛋白酶9(MMP9)表达的影响及其信号传导途径。方法 :明胶电泳酶谱分析法检测IL -1β对膀胱癌T24细胞MMP9表达的影响 ;蛋白印迹分析T24细胞丝裂原激活蛋白激酶 (MAPK)和磷酸肌醇3羟基激酶 (PI3K)传导通路在IL -1β作用下的磷酸化状态 ;用MAPK和PI3K传导通路的特异性抑制剂 ,对IL -1β 调控膀胱癌细胞MMP9表达的阻断研究。结果 :IL -1β 对膀胱癌T24细胞的MMP9表达有剂量依赖性调节作用 ;IL -1β 介导PI3K下游转录因子NF -κB时间依赖性激活 ;PI3K传导通路抑制剂Wortmannin阻断IL -1β 对MMP9表达上调作用。结论 :PI3K传导通路的激活介导IL -1β对膀胱癌T24细胞MMP9表达的上调作用。
Objective:To study the effect of interleukin-1β(IL-1β)on the expression of matrix metallpro-teinase9(MMP9)and signaling pathway of bladder carcinoma cells.Methods:The effect of IL-1βon the ex-pression of MMP9on bladder carcinoma cell line T24via zymograph was detected.Western blot was applied to test the phosphorylation of transcriptional factors downstream of MAPK and PI3K signaling pathway triggered by IL-1β,and attempted to block IL-1βregulated MMP9expression upon T24cells by MAPK and PI3K pathway specific inhibitors.Results:IL-1βregulated T24cells MMP9expression in dose-dependent manner.IL-1βreceptor transduced the timedependent activation of NF-κB through PI3K pathway.PI3K pathway specific inhibitor(Wortmannin)can block up-regulation of MMP9by IL-1β.Conclusion:IL-1βup-regu-lated MMP9expression through PI3K signaling pathway.
