牛磺酸对降低庆大霉素耳毒性作用的实验观察

摘要 目的探讨喂服一定剂量牛磺酸对豚鼠庆大霉素耳毒性的影响。方法 30只豚鼠随机分为3组,即庆大霉素组(GM 组):GM 80 mg/kg,腹腔注射;庆大霉素+牛磺酸组(GM+Tau 组):GM 80 mg/kg,腹腔注射,Tau 400 mg/kg,口服;对照组:生理盐水1 ml,腹腔注射。3组动物均每日给药一次,连续给药12 d。每组动物给药前及给药12 d 后分别测试旋转后眼震颤时间(post-rotatory nystagmus,PRN),听觉脑干诱发电位(auditorybrainstem response,ABR)的阈值、阈移及 click 不同声强度下Ⅲ波的幅值,12 d 测试后行耳蜗扫描电镜检查。结果(1)动物自身对照和组间对照显示,3组给药前后的 PRN 均无显著性变化。(2)给药12 d 后的 ABR 平均阈移:对照组为0.25~0.5 dB;GM 组 click 为8.3±5.2 dB,短纯音的平均阈移为3.3~18.5 dB,并以6、8 kHz 的阈移最为明显;GM+Tau 组 click 为0.5±1.5 dB,短纯音的平均阈移为0.5~2.0 dB,其中 click 及6、8 kHz 的平均阈移显著低于 GM 组的相应指标(P<0.05),而与对照组相比 click 及短纯音的 ABR 平均阈移差异均无显著性(P>0.05)。(3)各组给药12 d 后 clickⅢ波的幅值:GM+Tau 组与对照组十分接近,不同声强度引导的Ⅲ波幅值与对照组比较差异均无显著性;而 GM 组在各刺激强度下的Ⅲ波幅值均显著低于 GM+Tau 或对照组(P<0.05)。(4)耳蜗扫描电镜显示 GM 组底回损伤严重,GM+Tau 组损伤轻微。结论牛磺酸具有一定保护耳蜗抵抗庆大霉素耳毒性的作用。 Purpose To study the protective effect of taurine on gentamycin-induced ototoxicity in guinea pigs. Methods Thirty guinea pigs were randomly divided into three groups:GM group(GM 80 rng/kg,i.p.),GM+Tau group (GM 80 mg/kg,i.p.,Tau 400 mg/kg p.o.)and control group(saline lml,i.p.).All drugs were administered once a day for a consecutive 12 d.Before and 12 d after treatment,the duration of post-rotatory nystagmns(PRN),the thresholds of auditory brainstem response(ABR)and the amplitudes of wave Ⅲ after click were measured respectively.The threshold shifts were calculated afterwards.After the final test,some cochleas were prepared for scanning electron microscopy(SEM). Results(1)In intra-groups and intra-individuals,there was no significant difference in the duration of PRN before and after drug administration.(2)The mean threashold shifts of ABR 12 d after treatment was 0.25-0.5 dB in control,and 8.3± 5.2 dB of click and 3.3-18.5 dB of short tone in GM group with the most prominent at 6,8 kHz stimuli.For GM+Tau group,the mean threashold shifts were 0.5±0.5 dB after click,and 0.5-2.0 dB after short tones with the values of click and 6、8 kHz significantly lower than those of the GM group(P<0.05),but not when compared with the control group(P> 0.05).(3)The amplitudes of wave Ⅲ after click 12 d after treatinent were similar between GM+Tau and control group with no significant difference in amplitudes of wave Ⅲ after different sound intensity.For GM group,all the amplitudes at different sound intensity were significantly lower than those of the other two groups(P<0.05).(4)The SEM showed that the damage in GM group was severe but very slight in GM+Tau group.Conclusion Taurino has a potential protective effect against gentamycin-induced ototoxicity.
