摘要
目的:在正常和被链佐星诱导成糖尿病的大鼠体内,观察并比较海带对二甲双胍代谢的影响。方法:利用高效液相(HPLC)法测定给药后不同时间点在正常和被链佐星诱导成糖尿病的大鼠体内二甲双胍的血药浓度处理,药代动力学参数利用 RSTRIP 处理。结果:在正常和被链佐星诱导成糖尿病的大鼠体内,二甲双胍药代动力学参数相比较时,t_(1/2),AUC 及 MRT 有统计学上的差异;对糖尿病大鼠实验组,ka 及 AUC 也有统计学上的差异。但对血糖的影响没有显著的差异性变化。结论:海带对二甲双胍的个别药代动力学参数有改变,但对其他动力学参数及血糖的影响有待一步探讨。
Objective:To study the effect of Laminaria japonica diet on the absorption,distri- bution,metabolism and excretion of metformin which is frequently used in the treatment of dia- betes.Methods:The blood sample at different times after administration metformin to rats were collected.The plasma concentrations of metformin were determined by HPLC.The pharma- cokinetic parameters were accessed by RSTRIP.Results:Administration of metformin in strepto- zotocininduced diabetic rats treated with Laminaria japonica diet showed pharmacokinetic parame- ters similar to those without sea tangle diet.However,t_(1/2) Vss,AUC,MRT were increased com- pared to those of normal rats.This might result from blood glucose level responded to metformin concentration-independently,remaining almost constant over certain blood concentration of met- formin.Conclusion:More studies should be followed to fully understand pharmacokinetic changes of hypoglycemic agents caused by short and long-term Laminaria japonica diet.
