摘要
对复合物胶束PLL PEG PLL/DNA的形态、大小、对核酶抗性和转染效果等进行了测试,结果表明:复合物胶束直径在10~35nm之间,有较强核酶抗性,对昆虫细胞的转染实验获得成功。这些结果对于基因治疗具有重要意义。
As a non-viral vector in gene therapy,polycation homopolymer is cytotoxic and not biocompatible because of its positive charge nature.There will be insoluble aggregations when the stoichiometric charge ratio is 1.To eliminate such disadvantages,the hydrophilic polymer PEG was incorporated to form tri-block copolymer,which makes the cytotoxity of block copolymer decreased and the fluid increased and they wont aggregate even if at stoichiometric point.The shape,size,resistance to nuclease and transfection of micelles were measured after the formation of block copolymer complexes,and the results showed that the diameter of block copolymer/DNA micelles was between 10 to 35 nm; it was resistant to nuclease and the tansfection of insect cell-lines was successfully expressed,which might be significant important for gene therapy.
