摘要
试验研究了用高效液相色谱法检测替米考星在绵羊血清中的含量。绵羊血清用C18SPE柱净化,甲醇—乙腈—乙酰胺(20mL∶80mL∶0.77g,)洗脱,采用反相高效液相色谱测定,检测波长288nm。在血清中0.05,0.50,5.00mg(L-1添加回收率范围为92.3%～101.0%,变异系数为4.7%～8.0%。用该方法测得绵羊血清中替米考星含量的最低检测限为0.025mg(L-1,定量限为0.05mg(L-1。
A high performance liquid chromatography (HPLC) method was reported for the determination of tilmicosin in sheep serum.Sheep serum was purified by C(18) solid-phase extraction cartridges with methanol-acetonitrile-ammonium acetate (20 mL∶80 mL∶0.77 g), and the eluate was determined by HPLC at 288 nm. Recoveries were 92.3 %～101.0 % fortified at 0.05,0.50,5.00 mg ·L^(-1) in sheep serum.Coefficients of variation were in the range of 4.7%～8.0%.The minimum limit of (detection)(LOD) and limit of quantification (LOQ) with this method were 0.025 mg·L^(-1) and 0.05 mg·L^(-1) in serum respectively.
