摘要
目的:评价中国小型猪咽腭部注射聚丙烯酰胺水凝胶(hydrophilicpoly-acrylamidegel,HPG)制备睡眠呼吸暂停动物模型的可靠性、稳定性和安全性。方法:对10只中国小型猪咽腭部注射HPG,术后即刻及1周后分别观察动物血氧饱和度、睡眠呼吸暂停时间和次数,并对注射部位进行病理学观察。结果:采用HPG注射可造成明确的动物阻塞性呼吸睡眠暂停。术后即刻和术后1周后的平均呼吸暂停时间分别为(12.33±4.06),(53.94±6.51)s,比术前犤(6.63±0.89)s犦明显延长,差异有显著性意义(t=-6.137,-15.223,P<0.05)。术后即刻与术后1周呼吸暂停次数与术前比较显著增加(t=-7.048,-17.458,P<0.05);血氧饱和度显著下降(t=11.603,17.889,P<0.05)。病理结果显示HPG周围包膜完整,没有引起组织损伤和坏死。结论:HPG咽腭部注射可很好的诱导小型猪睡眠呼吸暂停,对周围组织没有造成损伤,所制备的模型具有可靠、稳定和安全的特点。
AIM:To evaluate the reliability, stability and safety of injection of hydrophilic polyacrylamide gel (HPG) to palatopharyngeal arch for establishing Chinese small swine model of obstructive sleep apnea. METHODS: HPG was injected into palatopharyngeal arch of 10 Chinese small swines to induce obstructive sleep apnea. The blood oxygen saturation, sleep apnea duration and number of times were observed immediately and 1 week after operation, and pathological observation to the injection site was performed. RESULTS: HPG injection could result in obstructive sleep apnea effectively. The average apnea duration immediately and 1 week after operation were (12.33±4.06)and(53.94±6.51) s respectively, which were significantly prolonged as compared with that before operation [(6.63±0.89) s] (t=-6.137,-15.223, P< 0.05).Compared with before operation, the times of apnea immediately and 1 week after operation were significantly increased (t=-7.048,-17.458,P< 0.05), but the blood oxygen saturation was significantly decreased (t=11.603,17.889,P< 0.05). The results of pathological observation showed that there was integrated integument around HPG, and no tissue damage and necrosis were observed. CONCLUSION: HPG injection can result in obstructive sleep apnea effectively without any tissue damage around the injected site. The established model is steady, reliable and safe.
