摘要
背景:近年来发现,脑内的星形胶质细胞能产生EPO,EPO含量升高可以阻抑急性缺血性神经细胞的损伤。目前EPO在慢性脑损伤中的变化和作用的文献报道较少。目的:研究慢性铝中毒对大鼠端脑和血清EPO水平及端脑皮质神经细胞的影响,探讨EPO在中药防治慢性铝中毒所致大鼠端脑皮质神经细胞形态学异常中的作用。设计:随机,对照的实验研究。地点和材料:实验地点:河北医科大学生物化学与分子生物学教研室。将雌性SD大鼠45只,随机等分为正常组、慢性铝中毒组和中药防治组。干预:用9g/L的AlCl3溶液对慢性铝中毒组和中药防治组大鼠腹腔注射,正常组大鼠腹腔注射等量生理盐水。对中药防治组大鼠灌胃中药,中药灌胃和铝盐造模同时进行,每次灌胃药液的生药含量是2.6g,1次/d,连续灌胃7d,间隔1d,共计72d;按上述周期和时间对正常组和慢性铝中毒组大鼠灌胃等量的自来水。从各组中随机抽取2只大鼠,常规切片和苏木精-伊红染色。各组中的其余大鼠,取端脑皮质和血清,用放射免疫法测定其中EPO含量。主要观察指标:大鼠端脑皮质组织细胞的变化,端脑皮质和血清EPO含量。结果:慢性铝中毒大鼠端脑皮质神经细胞出现多种病理性变化,端脑皮质EPO含量(0.13±0.01)μg/L降低(P<0.05),血清EPO含量(2.34±0.30)μg/L升高(P<0.05)。
BACKGROUND: Recently,it is discovered that erythropoietin(EPO) is also produced by astrocytes.The increase of cerebral EPO can protect neurons from acute ischemic damage.Effects of EPO in chronic cerebral damage have not been reported till now.OBJECTIVE:To study the impact of chronic Aluminum(Al) toxication on EPO levels in both telencephalon and serum as well as the neurocyte of cerebral cortex for the investigation of the effect of EPO on the prevention of abnormal cytomorphology of neurocyte in the cerebral cortex induced by chronic Aluminum toxication by Chinese herbs.DESIGN:A randomised controlled perspective study.SETTING and SUBJECTS:Department of Molecular Biology and Biochemistry of Hebei Medical University.Forty-five female SD rats were randomly allocated into normal control group(N group),chronic Al toxication group(M group),and Chinese herb prevention group(T group).INTERVENTIONS: 9 g/L of AlCl3 solution was injected into abdominal cavity of the animals in M and T group.Normal saline with same volume was injected into the animals of N group.Herbs were administrated through stomach-perfusion in the rats of T group.The herb perfusion and the modeling were operated simultaneously.The dose of raw herb was 2.6 g/each time and once a day for a continuous 7 days and with one-day interval for a total of 72 days.Tap water in the same volume was perfused into the rats of N and M groups according to the above medication perfusion cycle and time.Two rats were randomly selected from each group.Routine sectioning and HE staining were made.The cortex of cerebrain and serum of the rest rats of each group were taken for assay the EPO contents by radioimmunoassay. MAIN OUTCOME MEASURES:Pathological changes of cerebral neurons by HE staining, cerebral cortical and serum EPO level by radioimmunoassay.RESULTS:There were many pathological changes in cerebral cortical neurons of chronic Al toxication rats, the cortical EPO content in telencephalon decreased[(0.13±0.01) μg/L,P< 0.05],while the serum EPO content increased[(
