摘要
经筛选得到了一株能以原油为碳源进行生长繁殖的菌株 ,初步鉴定为芽孢杆菌 (L 32 ) ,对其发酵条件进行了优化 ,确定出最佳的发酵条件 ,并确定了厌氧条件下以原油为碳源时的培养基成分。通过补加葡萄糖使其浓度保持在8g/L的方法可以提高菌体的浓度。实验结果表明 ,L 2 3可以以原油为碳源生长 ,并能在降低原油粘度的同时使原油培养基内水相的pH值下降。L 2 3能提高实验岩心的原油采收率 ,即在关井 96h后 ,L 2 3可使原油采收率提高 9.6 %左右。
A piece of bacillus L-32, which is growing and procreating by taking crude oil as its carbon source, was screened. The fermentation condition to produce L-32 was optimized. The concentration of L-32 could be enhanced by adding glucose with a concentration of 0.8 percent in the medium. The medium for growing L-32 by taking crude oil as carbon source was optimized. The experiment demonstrated that L-32 could grow by taking crude oil as its carbon source and could reduce oil viscosity and produce some acid to lower pH value of culture medium. Under laboratory conditions, L-32 could increase oil recovery in a model reservoir by about 9.6 percent after oil well shut-in for 96 hours.
