摘要
动物细胞灌注培养过程中,活细胞的选择性截留是实现细胞高活性、高密度培养的关键。通过简单的数学模型对倾斜式沉降器中细胞沉降的过程进行分析,估算过程参数,并依据预实验作适当修正。杂交瘤细胞沉降实验结果表明,该沉降器对活细胞截留效率高于死细胞,具有截留选择性。所建过程模型对过程操作具有预见性,可用于过程参数的估计,并可指导沉降装置的设备放大。
Selective retention plays a critical role in the realization of animal cells perfusion culture with highviability and concentration. The down-flow settlement process in slanted cell settler was investigated through a simplifiedmathematical model. The values of relative parameters were estimated from the model computation, and received somemodification according to primary experiments. Results of controlled experiments with hybridoma cells indicated that theretention efficiency of viable cells was higher than dead cells in a selective mode. The established process model was ofvalue to the operation and the estimation of process parameters, and could facilitate the scale-up of cell settlement equipments.
