抑癌基因PTEN及P53在肝外胆管癌的表达 被引量:2

摘要 目的 :探讨PTEN及P5 3的表达与肝外胆管癌发生、发展的关系 ,两者表达的相关性及其临床意义。方法 :应用免疫组织化学SABC法检测 4 0例肝外胆管癌标本及 10例正常胆管组织PTEN及P5 3蛋白表达情况 ,应用RT PCR方法分析肝外胆管癌细胞P5 3mRNA的表达情况 ,同时结合临床资料进行分析。结果 :①肝外胆管癌中PTEN及P5 3蛋白的阳性表达率分别为 30 % (12 / 4 0 ) ,4 7.5 % (19/ 4 0 ) ,而正常胆管组织PTEN及P5 3蛋白的阳性表达率分别为 90 % (9/ 10 ) ,0 % (0 / 10 ) ;②两者的表达与肝外胆管癌部位无关 ,但均与胆管癌的病理分级相关 ;③PTEN的表达与胆管癌的转移情况相关 ,而P5 3的表达与转移情况无关 ;④PTEN与P5 3蛋白在肝外胆管癌中表达具有相关性 ;⑤本研究选用引物扩增出的P5 3cDNA为 4 31bp ,电泳结果与预期结果一致。结论 :PTEN蛋白在肝外胆管癌中低表达而P5 3蛋白在肝外胆管癌中高表达 ,提示PTEN及P5 3在肝外胆管癌的发生发展中起重要作用。P5 3突变可能下调PTEN的表达。 Objective: To evaluate the possible association of the expression of PTEN and P53 with the development and malignancy of extrahepatic cholangiocarcinoma (ECHC)and their clinical significance. Methods: The expression of PTEN and P53 in 40 cases of ECHC and 10 cases of normal choledochus were detected by method of immunohistochemistry. P53 mRNA expression was observed in ECHC cell line by reverse transcriptase polymerase chain reaction(RT-PCR). The results obtained were analyzed with clinical data. Results: By immunohistochemistry, PTEN and P53 protein positive expression rate was 30%(12/40) and 47.5%(19/40),respectively, whereas in 10 cases of normal choledochus the positive expression rate of PTEN and P53 was noted in 9(90%) and 0(0%) cases, respectively. Both the protein expression of PTEN and P53 gene had no relation with sex, age and tumor locus, but were related with the pathological grade. On the other hand, PTEN was related with the metastasis, while P53 was not. The PTEN gene had correlation with the P53 gene in protein expression. The length of P53 cDNA in ECHC cell line by RT-PCR was 1 431 bp which coincided with the expected. Conclusion: There exists abnormal expression of PTEN and P53 in ECHC. The expression of P53 gene may down-regulate the expression of PTEN gene.
