人骨髓间充质干细胞复合新型生物材料向软骨分化的实验研究 被引量:1

摘要 目的 探讨培养的人骨髓间充质干细胞体外经过软骨诱导分化植入体内形成软骨组织的方法。方法 分离培养人骨髓间充质干细胞 ,接种至新的高分子生物材料 3-羟基丁酸、3-羟基己酸无规嵌段共聚物p(3HB co 3HH)中 ,加入成软骨诱导培养体系 ,1周后植入裸鼠体内 ,10周后取出后行HE与阿尔新蓝染色。结果 HE与阿尔新蓝染色提示有软骨组织形成。结论 人骨髓间充质干细胞能够向软骨细胞分化 ,复合生物材料体内植入后能够构建软骨组织。 Objective In specific condition, mesenchymal stem cells (MSCs) can be differentiated along multiple mesenchymal lineages. A new method was investigated to reconstract cartilage. Methods The cultured human mesenchymal stem cells obtained from bone marrow were seeded onto the three-dimensional synthetic bioabsorbable polyester materials-p(3HB-co-3HH) manufactured by Qin Hua university . The compound was exposed to the differentiated liquid for chondrocytes lineage and then cultured for 1 weeksin vitro and embeded for 8 weeksin vivo. HE and Alcian blue staining were performed. Results The formation of cartilage was observed, and the polymer substrate support the MSCs proliferation. Conclusion Mesenchymal stem cells which seeded onto the synthetic bioabsorbable polyester materials can generate cartilage.
