摘要
目的:探讨苏芪合剂对脂多糖诱导的羊软骨细胞一氧化氮合成的调节作用,及其治疗类风湿关节炎(rheumatoidarthritis,RA)和骨关节炎的机制。方法:取健康小尾寒羊左后膝关节软骨,分离出软骨细胞进行体外培养。用脂多糖刺激软骨细胞分泌一氧化氮,同时加入不同浓度的苏芪合剂,分别于24,48和72h收集培养细胞上清液,用化学比色法测一氧化氮含量。结果:不同浓度的苏芪合剂对软骨细胞的一氧化氮合成均有抑制作用(P<0.01),且以剂量依赖方式。浓度为0.500g/mL的对软苏芪合剂骨细胞一氧化氮合成的抑制作用最强最稳定。结论:本实验证明了苏芪合剂抑制了软骨细胞一氧化氮的合成,可能是其治疗RA和骨关节炎的作用机制之一。
AIM:To study the regulation effect of suqi heji on nitric oxide(NO) synthesis in chondrocytes induced by lipopolysaccharide,and the pathogenesis of rheumatoid arthritis(RA) and osteoarthritis. METHODS:Cartilage was selected from left knee joint of healthy small tailed sheep and chondrocytes were isolated and cultured in vitro.They were stimulated with lipopolysaccharide to excrete NO. At the same time,suqi heji with different concentrations was added and the culture supernatants were collected at 24,48 and 72 hours,and the NO content was measured by chromatometry. RESULTS:Suqi heji with different concentrations had the inhibitive effect on NO synthesis of chondrocytes£¨P< 0.01£(c)in dose dependent manner.The inhibitive effect of suqi heji at 0.500 g/mL was the strongest and stablest.CONCLUSION:This experiment proves that suqi heji inhibits the NO synthesis of chondrocytes,which is one of the mechanisms for treatment of RA and osteoarthrosis.
