摘要
目的 研究中国日本血吸虫线粒体DNA的遗传变异。方法 试剂盒抽提我国11个地域株日本血吸虫基因组总DNA,以特异性引物对其线粒体NADH(还原型烟酰胺腺嘌呤二核苷酸)脱氢酶1(ND1)和细胞色素C氧化酶1(CO1)片段进行PCR扩增,PCR产物纯化后作SSCP分析。结果 SSCP结果共出现15种条带类型,其中NDl片段中出现7种条带类型,分别是湖北省5地2种;中国台湾、湖南、安徽与江西各为1种;四川、云南同为1种。CO1片段中出现8种条带类型,其中湖北境内5个地域株中有2种;中国台湾株、湖南、安徽与江西各为1种,四川、云南株各为1种。结论 我国各地域株日本血吸虫存在不同程度的遗传变异。
To study the genetic variations of the Chinese strain of Schistosoma japonicum, genomic DNA was isolated with kit, and the gene fragments of the mitochondrial NADH dehydrogenase (ND1) and cytochrome C oxidase (CO1) were amplified by PCR and sequenced. Then the PCR products were to be used to analyze the single-strand conformation polymorphism (SSCP) of each sample after purification. The results showed that there was only one band in each sample of the PCR products of NDl and CO1, and no variation in size among the PCR products was demonstrated. However, a lot of genetic variations in DNA SSCP among all samples were found.It concludes that there are different genetic variations among all the geographic strains of S. japonicum in China.
