PARP1在结直肠癌的表达及对SW620增殖与凋亡的影响

摘要 目的探究PARP1在结直肠癌的表达情况及其与癌临床病理特征的关系,以及结肠癌细胞株SW620、Lo Vo细胞PARP1的表达,分析其对结直肠癌细胞株SW620增殖与凋亡的影响。方法应用免疫组化检测39例结直肠癌组织及癌旁正常组织中PARP1的表达;实时荧光定量PCR(QPCR)检测结直肠癌细胞株SW620、Lo Vo、结肠正常黏膜细胞中PARP1的表达;转染PARP1基因sh RNA表达载体后,MTT检测细胞增殖,流式细胞术检测细胞凋亡情况,QPCR、Western blotting分别检测cyclin D1及Caspase-3的基因与蛋白的表达情况。结果PARP1在结直肠癌组织中的阳性表达率为71.8%,显著高于癌旁正常组织的20.5%(P<0.05)。PARP1在结肠癌细胞株SW620、Lo Vo的表达显著高于正常结肠黏膜细胞。转染PARP1基因sh RNA后,PARP1水平明显减低,SW620细胞增殖能力减弱,凋亡能力增强。结论 PARP1在结直肠癌中高表达,可促进结直肠癌细胞增殖,抑制细胞凋亡。 Objective To investigate PARP1 expression in tumor and adjacent normal tissues from colorectal cancer(CRC)patients, and colon cancer lines SW620 and Lo Vo, and analyze the relationship between PARP1 expression and clinic pathologic features, and the effect of PARP1 expression on proliferation and apoptosis of SW620 cells. Methods Immunohistochemistry and q PCR were respectively used to detect PARP1 expression in CRC tissues and adjacent normal intestinal mucosa, and in SW620 and Lo Vo cells. After transfection with PARP1-sh RNA, the proliferation and apoptosis of SW620 cells were detected by MTT and flow cytometry. The gene and protein expressions of cyclin D1 and Caspase-3 were detected by q PCR and Western blotting. Results The positive expression rate of PARP1 was 71.8% in CRC tissues, significantly higher than 20.5% in adjacent normal intestinal mucosa tissues(P < 0.05). The expression levels of PARP1 in SW620 、Lo Vo cells were significantly higher than that in normal intestinal mucosa cells. After transfection with PARP1-sh RNA, PARP1 expression and cell proliferation were significantly inhibited, while tumor cell apoptosis increased. Conclusion PARP1 expression is high in CRC, which can promote proliferation and prohibit apoptosis of tumor cells.
