子痫前期患者胎盘中miRNA-101和内质网蛋白ERp44的表达和意义 被引量:7

miRNA-101 regulates apoptosis of trophoblast cells in preeclampsia by targeting endoplasmic reticulum protein 44

摘要 目的探讨子痫前期患者胎盘中miRNA-101和内质网蛋白ERp44的表达和意义。方法选取2013年5月~2014年5月入住山东省交通医院分娩的100例产妇的胎盘绒毛组织,随机均分为对照组和子痫前期组,每组各50例。采用RT-PCR法检测各组胎盘组织中miRNA-101的表达情况;Western blot法检测过表达和封闭miRNA-101后,各胎盘组织ERp44的表达情况;采用流式细胞仪检测滋养细胞的凋亡情况。结果与对照组相比,子痫前期组中miRNA-101的表达明显下调,差异有统计学意义(P<0.05)。在滋养细胞HTR-8/SVneo中过表达miRNA-101后,ERp44的表达显著下调(P<0.05);封闭miRNA-101后,ERp44的表达上调(P<0.05);过表达miRNA-101,细胞凋亡数量减少;封闭miRNA-101,细胞凋亡数量增加。结论子痫前期中,miRNA-101可通过调控内质网蛋白ERp44的表达来调控胎盘滋养细胞的凋亡过程。 Objective To explore the possible mechanism of miRNA-101 regulates endoplasmic reticulum protein ERp44 and apoptosis of placental trophoblast in preeclampsia. Methods 100 cases experimental object from May 2013 to May 2014 in our hospital was selected and divided into two groups. The miRNA-101 expression in two groups were detected by RT-PCR. ERp44 expression were detected by western blot when overexpressed and inhibited miRNA-101. At the same time,the number of cell apoptosis was tested by Flow cytometry. Results Compared with control group,the expression of miRNA-101 in PE group were decreases( P < 0. 05). The level of ERp44 was elevated and the number of cell apoptosis reduced after miRNA-101 overexpression; the level of ERp44 was declined and the number of cell apoptosis increased after inhibite the expression of miRNA-101( P <0. 05). Conclusion mi RNA-101 regulates apoptosis of trophoblast cells by regulating the expression of ERp44 in patients with preeclampsia.
