防冻剂种类以及投入液氮时温度对昆明小鼠胚胎冷冻保存效果的影响

摘要 分别以2mol/L的甘油,二甲基亚砜和乙二醇做防冻剂对昆明小鼠桑椹胚和早期囊胚进行冷冻保存,并在同剂量防冻剂中分别添加了0.5mol/L蔗糖进行对照实验;将胚胎在-20、-25、-30℃投入液氮,观察其对胚胎冷冻效果的影响。结果表明:2mol/L乙二醇和2mol/L甘油+0.5mol/L蔗糖处理组冷冻效果最好;在-30℃时投入液氮的胚胎成活率最高;将解冻后的胚胎移植给7只受体雌鼠,共得到26只仔鼠。 The morulae and early blastocysts of kun Ming strain mouse were frozen by using 2mol/L glycerol dimithyl sulfoxide and glycol as the cryoprotectants in this study and 0.5mol/L sucrose were supplemented to each of them as control groups.After cooled the embryos were plunged into liquid nitrogen at different temperatures and the influence of these factors to the freezing effects were observed. The results showed that 2mol/L glycol and 2mol/L glycero1+0.5mol/L sucrose had the best freezing effect.The high survival rate was obtained after culturing the thawed embryos which were plunged into liquid nitrogen at 30℃. The frozen- thawed embryos were transferred to 7 receipient mice and 26 newborns were obtaind.
