摘要
本实验观察了硒代二半胱氨酸对白血病细胞系(U937和K562)第二信使的影响,旨在探讨其抑制白血病细胞生长和有丝分裂的机制。结果表明,经3.0μmol/L(半数抑制浓度)的硒代二半胱氨酸作用3天后,白血病细胞内cGMP和Ca^(++)含量明显降低,cAMP含量显著增高,CAMP/cGMP比值增大,从而抑制白血病细胞DNA合成期所需的胸腺嘧啶核苷酸合成酶活性,抑制有丝分裂期微管蛋白的解聚,干扰有丝分裂过程,造成白血病细胞生长和增殖障碍。
The effects of dicysteine selenium on the level of cAMP, cGMP and Ca++in leukemic cell lines (U937 and K562) were observed. After incubated for 3 days with dicysteine selenium at the concentration of 3.0μmol/L, the levels of cGMP and Ca++decreased signigicantly (P<0.05, P<0.01) and the level of cAMP increased markedly(P<0.01) as compared with those of the controls. It is demonstrated that the dicysteine selenium may be a modulator of the differentiation and proliferation of leukemic cells, it could inhibit DNA systhesis and division of U937 and K562 leukemic cells with the high level of cAMP / cGMP ratio by reducing the cGMP and Ca++level, and increasing the cAMP level.
