摘要
目的:研究梁金菇多糖(LJPS)对造血功能的影响。方法:采用粒细胞巨噬细胞集落形成单位(CFU-GM)琼脂半固体培养法观察 LJPS 刺激的脐血单个核细胞(CBMC)上清液对脐血 CFU-GM 形成的作用,并采用逆转录聚合酶链反应(RT-PCR)的方法,从 mRNA 水平对其机理进行探讨。结果:LJPS 刺激的脐血单个核细胞上清液可促进脐血中 CFU-GM 的生成,最佳浓度为1μg/ml,与空白组比较差异有显著性(P< 0.01)。LJPS 对 CFU-GM 的生成没有直接作用,但可诱导 CBMC 表达 GM-CSF mRNA。结论:LJPS 具有促造血的功能。
To study the effect of Liangjin mushroom polysaccharide (LIPS)on hematopoietic function.Methods:Colony forming u- nit of granulocyte and macrophage(CFU-GM)agar sinai-solid cultivation were used to observe the effect of LIPS stimulated cord blood mononuclear cell(CBMC)supernatant on CFU-GM formation,and reverse transcription polymerase chain reaction(RT-PCR)was used to study the mechanism on mRNA level.Results:The supernatant of LIPS stimulated CBMC could improve the CFU-GM forma- tion,the optimal concentration was 1 μg/ml,and has significant difference in comparing with the control group(P<0.01).LJPS showed no direct effect on the growth of CFU-GM,but can induce the expression of granulocyte and macrophage colony stimulating factor(GM-CSF)mRNA.Conclusion:LIPS can promote hematopoietic function.
