摘要 胶质瘤常规的治疗方法如手术、放疗和化疗都有不尽人意的地方,免疫治疗已成为肿瘤的第4种治疗模式,CIK细胞的过继免疫治疗可望成为一种有前途的辅助治疗方法。本实验观察胶质瘤患者细胞因子诱导的杀伤细胞(CIK)的特性,CIK细胞对胶质瘤的体外杀伤活性。方法:分离胶质瘤患者的外周血单个核细胞(PBMCs),加入不同的细胞因子,体外培养成CIK细胞;用流式细胞仪对CIK细胞进行动态表型分析;采用MTT法测定胶质瘤患者及正常人CIK细胞体外细胞毒活性。结果:CD3^+/CD56^+双阳性细胞迅速增多,由起初的3.45%上升到第30天的55.2%;CIK细胞对肿瘤细胞的杀伤活性在一定程度上与效靶比呈数量依赖关系、培养第18天左右杀伤活力达高峰,CIK细胞的杀伤活性高于LAK细胞,胶质瘤患者CIK细胞杀伤活力较正常人低下。结论:CIK细胞对胶质瘤具有较好的杀伤活性,较LAK细胞更适于临床治疗。 BACKGROUD & OBJECTIVE: Immunotherapy has become one of the most important alterna-tive therapies for glioma patients. This study was designed to determine the characteristic of the antitumor activity of cy-tokine-induced killer (CIK) cells derived from glioma patients. METHODS: Peripheral blood mononuclear cells(PBMCs) from glioma patients were incubated in vitro and induced into CIK cells in the presence of various cytokines.PBMCs from health donor were used as control The phenotype of CIK cells were identified by flow cytometric analysis.The cytotoxicity of CIK cells against glioma cells was determined by MTT assay. Cytotoxicity of CIK cells were compared with that of lymphokine activated killer (LAK) cells. RESULTS: The percentage of CD3^+ /CD56^+ positive cells was signifi-cantly increased from 3. 45% on day 0 to 55. 2% on day 30 of incubation. With the increased ratio of effector and tar-get cells, the cytotoxic effect of CIK cells correspondingly became stronger. The highest antitumor activity of the CIK cells was on 18th incubation day. Cytotoxic effect of the CIK cells was significantly higher than that of the LAK cells.However CIK cells originated from glioma patients possessed a lower antitumor activity than that from health donor. CON-CLUSION: CIK cells, as compared with LAK cells, are highly effective cytotoxic cells against glioma and might be used as an alternative adoptive immunotherapy for glioma patients.
