低分子肝素对裸鼠原位胃癌生长及人端粒酶逆转录酶表达的影响 被引量:1

摘要 目的探讨低分子肝素(LMWH)的抗癌机制。方法应用原位移植方法建立裸鼠胃癌原位模型并随机分为4组各10只,术后1周5-氟尿嘧啶(5-Fu)组、对照组分别腹腔注射5-Fu30mg/kg及等量生理盐水,LMWH组皮下注射LMWH1500IU/kg),联合组按上述方法同时予5-Fu及LMWH,均为每周2次,共5周。第6周处死动物,测定裸鼠原位胃癌肿瘤体积、肿瘤增长抑制率(TGI),并采用RT-PCR法测定hTERT mRNA表达(OD值)。结果5-Fu组、LMWH组和联合组肿瘤体积明显小于对照组(P<0.05),联合组体积显著小于5-Fu组(P<0.05);LMWH组和联合组hTERT mRNA表达量均显著低于对照组和5-Fu组低(P均<0.05)。结论LMWH可抑制裸鼠原位胃癌瘤体生长并与5-Fu有协同作用,其机制可能与降低肿瘤hTERT mRNA的表达有关。
