摘要
本文用RT-PCR的方法,首次从草莓幼果RNA中成功扩增出KUP/HAK/KT转运体基因的部分序列,经过克隆测序得到了1个长度为1 252bp基因片段,编码416个氨基酸。经过与其他物种KUP/HAK/KT转运体基因序列比较相似率在70%~78%之间,氨基酸相似性在72%~88%之间。草莓KUP/HAK/KT转运体基因的成功克隆,为草莓抗褐变的研究及草莓钾元素营养代谢分子水平上的研究及果实品质的改良打下坚实的理论基础。
By using RT-PCR method,a partial sequences of Potassium transporter genes was amplified from cDNA of strawberry young fruit.After cloning and sequences,A sequence ranging 1252bp was obtained and encoded 416 amino acid.Compared the sequences with other Potassium transporter genes from Gene Bank,It reveals a similarity higher than 70%at the nucleotide level,and 72% at amino acid level.The success of cloning Potassium transporter gene fiom strawberry builds a great base for researching at the molecular level of Potassium nutrition metabolism and quality of fruit improving researches.
