摘要
目的 比较基因芯片与聚合酶链反应 (PCR)在人乳头瘤病毒 (HPV)检测分型中的优缺点。方法 抽提待检样品的核酸 ,扩增及荧光标记后与HPV检测及分型基因芯片杂交 ,用GMS 4 18扫描仪对杂交后的芯片进行扫描检测 ;同时用PCR的方法对这些样品进行HPV检测。结果 3例假性湿疣均无HPV检出 ,2 2例尖锐湿疣和 6例宫颈癌组织HPV均为阳性 ,4例CINⅡ~Ⅲ级中 3例HPV阳性 ,1例阴性 ;基因芯片检测结果与PCR结果一致。结论 HPV检测分型芯片是一种敏感高效的HPV检测分型方法。
Objective To compare the advantages and disadvantages of human papillomavirus DNA microarray and PCR in the detection and genotyping of HPV. Methods The samples amplified and labled with the fluorescence by PCR were hybridized with the HPV DNA microarray. The chips were scanned by GMS 418 Array Scanner.HPV was also detected by PCR in the samples. Results It showed HPV negative in all the 3 pseudocondyloma;HPV positive in all the 26 condyloma acuminata and 6 cervical cancer samples;3 HPV positive and 1 negative in the 4 CINⅡ-Ⅲ samples. The results of gene chips were consistant with the results of PCR. Conclusion HPV DNA microarray is a sensitive and efficient way to HPV detection and genotyping.
