摘要
以一株枯草芽孢杆菌(Bacillus subtilis)菌株为材料,发酵生产环糊精葡萄糖基转移酶(CGTase)。采用单因素试验考察碳源、氮源、培养温度、初始pH等对酶活力影响,在此基础上设计正交试验优化发酵条件。结果表明,优化的发酵条件为以玉米淀粉作碳源、大豆粉作氮源、pH 6.5、30℃发酵培养36 h,菌株所产CGTase的酶活力可达5 046 U/mL。
Cyclodextrin glycosyltransferase(CGTase) was fermented by Bacillus subtilis.Orthogonal design based on single factor tests was applied to study the effects of carbon source,nitrogen source,culture temperature and initial pH on enzyme activity.Results showed that the optimum fermenting conditions were using maize starch as carbon source,soy flour as nitrogen source,fermenting at 30 ℃ with initial pH at 6.5 for 36 h.The CGTase activity could be up to 5 046 U/mL under these conditions.
