摘要
目的:研究Smad泛素化调节因子1(Smad ubiquitin regulatory factor-1,Smurf1)与骨形态发生蛋白-2(bone morphogenetic protein-2,BMP-2)在骨的良性病变及骨肉瘤组织中的表达及相关性,以此分析Smurf1蛋白在骨肉瘤的发生、侵袭、转移中的意义。方法:共收集骨组织标本120例,其中良性病变36例,骨肉瘤84例。免疫组化法检测标本中Smurf1和BMP-2含量,相关性分析采用Spearman等级相关分析法。结果:骨肉瘤组织Smurf1蛋白阳性表达明显高于良性病变组织[(0.786±0.214)vs.(0.583±0.183)],BMP-2蛋白表达水平则相反[(103.57±13.23)vs.(152.19±19.42)],差异均有统计学意义(P<0.05)。Smurf1与BMP-2在骨肉瘤组织中的蛋白表达水平呈显著负相关(r=-0.598,P<0.05)。结论:Smurf1可通过抑制BMP-2的表达来促进骨肉瘤的形成和发展。
Objective To study the expressions of Smurf1 and BMP-2 in bone osteosarcoma and benign lesion tissues,and then analyze their correlation and possible roles in the development,invasion and metastasis of osteosarcoma.Methods Total 120 cases of bone specimens were collected,36 cases of specimens were from benign lesion tissues,while the other 84 cases were from osteosarcoma.The protein expressions of Smurf1 and BMP-2 were detected with immunohistochemistry.The correlation between them was analyzed with Spearman analysis method.Results The positive expression of Smurf1 in osteosarcoma was significantly higher than that of benign lesion tissues[(0.786±0.214)vs.(0.583±0.183)],the expression of BMP-2 was exactly opposite[(103.57±13.23)vs.(152.19±19.42)],(both P<0.05).The expression of Smurf1 and BMP-2 in osteosarcoma was negatively correlated(r=-0.598,P<0.05).Conclusion Smurf1 can promote the development of osteosarcomas by inhibiting the expression of BMP-2.
