PGC-1基因Gly 482 Ser多态与2型糖尿病的关系

摘要 [目的 ]探讨过氧化物酶增殖体激活受体γ共激活蛋白 1(peroxisomeproliferator activatedreceptor γcoactivator 1,PGC 1)基因Gly 4 82Ser多态在中国延边地区汉族和朝鲜族群体中的基因型和等位基因频率的分布特征及与 2型糖尿病发生的关系 .[方法 ]采用聚合酶链反应 限制性片段长度多态性技术检测PGC 1基因编码区Gly 4 82Ser位点的多态性 ,并应用病例 对照法研究糖耐量正常组汉族39名、朝鲜族 36名 ,2型糖尿病组汉族 2 0名、朝鲜族 2 7名 .[结果 ]汉族糖耐量正常组与朝鲜族糖耐量正常组的A等位基因频率分别为 4 8 72 % ,4 3 0 6 % ,两者间无显著性差异 ;汉族糖尿病组与朝鲜族糖尿病组的A等位基因频率分别为 4 7 5 0 % ,4 0 74 % ,两民族糖尿病组与糖耐量正常组间的等位基因频率间均无显著性差异 ;在朝鲜族糖尿病组中 ,GG组的空腹血糖浓度低于GA/AA组 .[结论 ]PGC 1基因Gly 4 82Ser多态在中国延边地区汉族及朝鲜族群体中的分布无差异 ;在中国延边地区的汉族及朝鲜族群体中 ,Gly 4 82Ser多态与 2型糖尿病的发病无相关性 ,但在朝鲜族糖尿病组中 ,突变型等位基因A与患者的空腹血糖浓度水平相关 . OBJECTIVE?To determine frequencies distri bu tion of peroxisome proliferator-activated receptor-γ coactivator-1(PGC-1) gene Gly?482?Ser polymorphism genotype and allele in Korean-Chinese nationality and Han nationality of Yanbian area in China, and relation ship with type 2 diabetes mellitus(DM).?METHODS? Polymerase chain reaction-restriction fragment length polymorphism was used to test a cod ing variant Gly?482?Ser of PGC-1 gene in 39 Han nationality and 36 Korea n-C hinese nationality with normal glucose tolerant(NGT) and 20 Han nationality p atients and 27 Korean-Chinese nationality patients with type 2 DM. Case-cont rast analysis was used to study the association between Gly?482?Ser poly morp hism an d type 2 DM.? RESULTS?The A allele frequencies of NGT subje cts were 48.72% for Han nationality, 43.06% for Korean-Chinese nationality, and no significant difference was found between two groups. The A allele frequenci es of type 2 DM patients were 47.50% for Han nationality, 40.74% for Korean-Ch inese nationality. Allele frequency distribution of each ethnic was compared between DM group and NGT group. There were no significant differen ces. In Korean-Chinese nationality type 2 DM group, fasting plasma glucose (FPG) levels in GG group were lower than those in GA/AA genotype.?CONCLUSION?There was no difference in frequencies distribution of PGC-1 gene Gly?482?Ser polymorph ism between Han nationality and Korean-Chinese nationality of Yanbian area in China.The Gly?482?Ser polymorphism was not associated with type 2 DM in Han nationality or Korean-Chinese nationality of Y anbian area in China, but the mutated A allele was likely associated with FPG le vels of Korean-Chinese nationality type 2 DM patients.
